Validation of the mRNA expression levels determined with the microarray by quantitative real-time RT-PCR and correlation of the data. A) Individual values for mRNA expression by quantitative real-time RT-PCR. The real-time RT-PCR data are expressed as the log2 ratio of the fold change in the expression of each target gene using the relative quantification method by comparison with the expression of the housekeeping gene RPLP0 (Pfaffl, 2001). Each circle represents the mean of two replicates of the fold change between of each the obese sample and the pool of nonobese control samples (n = 12). B). Linear regression analysis of the results determined by microarray and real-time RT-PCR analyses. The x-axis represents the log2 ratios of the relative mRNA expression determined by real-time RT-PCR and the y-axis represents the log2 ratio obtained by microarray for each sample and gene analyzed (n = 167).
